چکیده

In this work the α and β-amylase enzymes were obtained from maize (Zea mays) malt and were biochemistry characterized. A germination study to obtain the maize malt with good amylase activity was made. The maize seeds were germinated in laboratory and the enzymatic activity was measured daily. Activity dependence to germination time were fitted to an exponential model (A=A0e ), which showed that the behaviour of enzymatic activity in the germination process was similar to the growth of the microorganism. Its model could be applied to describe the mechanism of α-amylase production for each maize varieties and others cereals. Maize malt characterization showed that α and β-amylase had optimal pH between 4-6.5, optimal temperature 50 and 90°C, and molecular weight of 67.4 and 47.5kDa, respectively. This work contributed with the advances in biotechnology generating of conditions for application of a new and of low price amylases source.

Characterization and expression of an antifungal zeamatin-like protein (Zlp) gene from Zea mays.

A cDNA clone encoding a basic thaumatin-like protein of Zea mays was recovered from a mid-development seed cDNA library. The gene, Zlp, encoded a protein that was nearly identical with maize zeamatin and alpha-amylase/trypsin inhibitor. Expression of Zlp mRNA was highest in the endosperm tissue of seed 4 weeks after pollination. Expression of zeamatin-like (ZLP) protein correlated with mRNA; al...

Statistical Optimization and Partial Characterization of Amylases Produced by Halotolerant Penicillium Sp

The production of extracellular amylase by the halotolerant fungus Penicillium sp. was statistically optimized in a submerged cultivation. Maximum enzyme production was obtained after two days of incubation in a medium containing 10% (w/v) NaCl, in addition to MgSO .7H O 1% and 1% (w/v) of 4 2 starch. The optimum pH for growth and enzyme production was 4. Partial characterization of the crude e...

جداسازی و شناسایی باسیلوس مولد آلفا آمیلاز گرمادوست: بهینه سازی شرایط تولید و بررسی فعالیت و پایداری دمایی

Alpha-amylases are the most important amylases used in industry. Among them, thermophilic alpha-amylases are of particular importance, which is due to their activity and stability in high temperatures. These enzymes produced by thermophile micro-organisms including bacteria. These thermophilic alpha-amylases are used in various industries such as processing of starch as well as production of de...

Enzymic dissociation of zea shoot cell wall polysaccharides : I. Preliminary characterization of the water-insoluble fraction of zea shoot cell walls.

The water-insoluble fraction, obtained after successive treatment of an insoluble fraction of a buffer-homogenate of Zea mays L. hybrid B73 x Mo 17 shoots with 3 molar LiCl and hot water, was treated with alpha-amylase to remove starch. This fraction has been subjected to sugar composition analysis and the major glycosidic linkages were determined. The results suggest that the water-insoluble f...
